Subcutaneous injection of 615 adult mice can induce leukemia with a median survival time Is 29.7 days. The spleens of the diseased mice were used as the tumor source, and 615 mice were subcultured and passaged. The disease is 100%, and the survival time gradually decreases. After more than 30 years of age, a stable leukemia model was established, called the L615 leukemia animal model.

      [Modeling method] After passing L615 leukemia cells in vitro, it is determined that the number of live cells accounts for 96% or more of the total number of cells, and 1×1000000 cells are injected into normal L615 mice through the abdominal cavity. .. Starting from the third day after vaccination, peripheral blood white blood cell counts, blood smears were removed, blood was collected every day, Wright's staining and peroxidase staining were used to check leukemia cells, which confirmed that the mice were infected and successfully established a model . Before death on the 7th day, the animal died of cervical dislocation before death, and after soaking in 75% ethanol for 10 minutes, the spleen was removed aseptically, the spleen was separated, and washed twice with pre-cooled PBS. Add the RPMI 1640 solution dropwise to the 100 mesh metal mesh until the fragmentation is complete. After the fragmentation is complete, collect the cell suspension and centrifuge at 1500r/min for 5 minutes. Discard the supernatant and discard the prepared pretreatment Treat with cold red blood cell lysate for 10 minutes. Re-filtered through 200 mesh nylon mesh, resuspended in RPMI1640 medium, counted, and inoculated 1×100,000 cells into the abdominal cavity of experimental L615 mice to obtain an animal model of T lymphocyte leukemia.

       [Model Features] The survival period of L615 leukemia mice is (6.7±1.2) days. In addition to subcutaneous vaccination, intraperitoneal vaccination can also cause 100% of diseases. The survival time is slightly shorter, but no ascites is formed. L615 mouse leukemia has different sensitivity to various anti-tumor drugs.
