Objective: To optimize the two-step chemical induction method and establish a BALB/c mouse skin cancer model.
Method: BALB/c shaved the mouse back to expose the skin of approximately 2 cm x 2 cm. The mice were randomly divided into a blank control group and four model groups. I used 1, 2 and 4 to draw 4 model groups. In the first week, apply 100μg/100μLDBBA/acetone solution 7 times a week, and then apply 4μg/100μLTPA/acetone solution twice a week to record the body weight, tumor occurrence time, tumor count, and sacrifice the mice 12 weeks later . For tumors, the tissue adjacent to the tumor and the tissue adjacent to the tumor were analyzed by HE staining.
Result: weekly administration of DMBA can prolong the tumor cycle. Twice a week, tumors have a short cycle and high quality. It damages the skin four times a week, but does not affect tumor formation. It caused great damage to the skin seven times a week and caused the death of mice.
Conclusion: DMBA was applied twice in the first week, and then TPA was continuously introduced to establish a BALB/c mouse skin cancer model. It is easy to operate, the tumor occurrence period is short, and the incidence of cancer is high. It is similar to the development and development of human skin cancer. Establish an ideal experimental animal model for skin cancer research.
