【Animal Modeling】-Improvement and evaluation of rat myocardial ischemia reperfusion injury model

  OBJECTIVE: To improve the preparation of rat acute myocardial ischemia-reperfusion injury (MI/RI) model and conduct a comprehensive evaluation to lay a good model foundation for subsequent MI/RI research.

  Methods: 36 clean Wistar rats were randomly divided into 3 groups: normal group (normal group), left anterior descending artery (LAD) without intervention, sham operation group (sham group), LAD with only threading and no ligation: ischemia In the reperfusion group (MI/RI group), the LAD was ligated for 30 minutes and reperfused for 120 minutes. After the rats were anesthetized, the electrocardiogram was recorded continuously, a ventilator was used for tracheotomy, and the rat’s LAD was involved in the thoracotomy. Improved electrocardiogram, measurement of myocardial enzyme content, myocardial infarction area and myocardial histopathological examination. Model evaluation.

  Results: The ST-T dynamic changes of the ECG in the MI/RI group were obvious; the creatine kinase isoenzyme (CK? MB) level of the MI/RI group increased significantly after 2 hours; compared with this group, the MI/RI group The levels of creatine kinase isoenzymes were significantly increased. The infarct size increased significantly, the cardiomyocytes in the MI/RI group were degeneration and necrosis, and the inflammatory cells infiltrated extensively.

  Conclusion: The improved modeling method not only reduces the difficulty of surgery, but also successfully establishes a rat MI/RI model, laying a good model foundation for future MI/RI research.
