【Animal Modeling】-Preliminary study on serum cytokine expression profile of collagen-induced arthritis/syndrome model rats

  Objective: To study the expression of serum cytokines in collagen-induced rheumatoid arthritis, cold-damp arthralgia and damp-heat arthralgia/syndrome model rats.

  Methods: The model of rheumatoid arthritis (RA) was replicated with bovine type II collagen and Freund's complete adjuvant, and on this basis, cold-dampness and damp-heat stimulation were added to establish a model of combining cold-dampness arthralgia and damp-heat arthralgia syndrome. Array antibody chip detects the expression of serum cytokines.

  Results: Compared with the normal group, the RA group activinA, CNTF, IFN-γ, TNF-α up-regulated, MIP-3α, MMP-8 down-regulated; Hanshibi group CINC-2α, GM-CSF, IFN-γ up-regulated, MIP- 3α is down-regulated; ICAM-1 is up-regulated in the damp-heat arthralgia group, IFN-γ, MIP-3α, RAGE is down-regulated. Compared with the RA group, β-NGF, CINC-2α, MMP-8, PDGF-AA is up-regulated, and activin is in the Hanshibi group. TNF-α is down-regulated; MMP-8 is up-regulated in the damp-heat arthralgia group, activin, CNTF, IFN-γ, IL-1α, IL-6, MIP-3α, RAGE, TNF-α is down-regulated. The cold-damp arthralgia group is compared with RA Rebi damp-heat Compared with the Bi group, CINC-2α, IFN-γ, IL-1α, MIP-3α, PDGF-AA, and RAGE were all up-regulated. Compared with the normal group, the ratio of IFN-γ/IL-4 in the RA group and the Hanshibi group increased , The ratio of damp-heat arthralgia group was significantly reduced; compared with RA group, the ratio of cold dampness arthralgia group increased, and the ratio of damp-heat arthralgia group decreased significantly.

  Conclusion: Disease/syndrome factors have a significant impact on the serum cytokine expression profile of rats, and there are significant differences between the models.
