Objective: To observe the effect of electroacupuncture on p65 protein in the posterior horn of spinal cord in rats with diabetic neuralgia, and to explore the local analgesic effect of electroacupuncture on diabetic neuralgia.
Method: 30 SD rats were divided into normal group and model by completely random grouping method, and were given a normal diet and a high-fat/high-sugar diet for 5 weeks. After feeding for 5 weeks, the model rats were injected intraperitoneally with streptozotocin (STZ), and then randomly divided into model group (DNP group) and model + electroacupuncture group (DNP + EAGrup). The rat makes a selection. I divided it into two and chose the double point. "Zunsanri, Kunrun Point", frequency 2Hz, intensity 1mA intervention for 15 minutes, 2mA intervention for 15 minutes, once a day, 7 times from the 7th week. Observe the changes in blood glucose, mechanical pain threshold and thermal pain threshold of each group of rats. Hematoxylin-eosin (HE) staining was used to observe the morphological changes of rat sciatic nerve, and the posterior horn of rat spinal cord was detected by western blotting. . Result: After feeding high-fat and high-sugar for 5 weeks, rats were injected with STZ intraperitoneally for 2 weeks. Blood glucose levels increase, and mechanical and thermal pain thresholds decrease. This is because DNP was successfully established and electroacupuncture can up-regulate DNP rats. The mechanical and thermal pain thresholds of DNP have no obvious intervention effect on blood glucose. The results of HE staining showed that the myelinated nerve fibers of the sciatic nerve of DNP rats were randomly arranged. Axial swelling, heterogeneous myelin density, vacuole degeneration, and electroacupuncture intervention on the sciatic nerve morphology of DNP rats had no significant effect. Western blot results showed that the expression of NF-κBp65 protein in the spinal dorsal horn of DNP rats increased significantly. , Electroacupuncture may down-regulate the expression of NF-κBp65 protein in the spinal dorsal horn of DNP rats.
Conclusion: Low-frequency electroacupuncture has a good analgesic effect in DNP model rats, which may be related to the inhibition of the expression of NF-κBp65 protein in the spinal dorsal horn of diabetic DNP rats.
