【Animal Modeling】-miRNA-144 gene overexpression has antitumor effect on liver cancer mice and its influence on intestinal flora

Objective: To analyze the anti-tumor effect of microRNA-144 (miRNA-144) gene overexpression on mice with liver cancer and its influence on the intestinal flora. How to establish ascites liver cancer transplanted into mice? Observe the effect of overexpression of miRNA-144 gene on the general condition of liver cancer mice, the histopathological morphology of liver and intestine, the richness and diversity of intestinal flora, and the intestinal barrier function. Real-time fluorescent quantitative polymerase chain reaction and Western blotting were used to determine the status of occludin, ZO-1, miRNA-144 mRNA, occludin and ZO-1 protein.

Results: The overexpression of miRNA-144 improved the overall physical condition of liver cancer mice, inhibited the development of liver cancer, improved the intestinal microeconomic environment and intestinal mucosal barrier function, colon tissue occluder, ZO can up-regulate -1, The expression of miRNA-144 mRNA, occludin and ZO-1 protein.

Conclusion The overexpression of miRNA-144 may inhibit the development of primary liver cancer and improve the intestinal microenvironment and intestinal mucosal function of mice. This mechanism may be related to the up-regulation of occludin, ZO-1 mRNA and protein expression.
