[Disease animal model]-rat model of high altitude polycythemia

  Altitude sickness (called "altitude sickness") is a chronic altitude sickness caused by excessive red blood cell proliferation (excessive red blood cell proliferation) caused by high altitude hypoxia. Compared with the same highly healthy person, the number of red blood cells, hemoglobin and red blood cells in patients with high red blood cells is significantly increased, and the arterial blood oxygen saturation is reduced, accompanied by excessive clinical symptoms and signs. The disease is common in high-altitude immigrants, but it is rare in high-altitude people. The incidence rate in men is significantly higher than that in women, and there are few cases in children. Hypoxia, ultra-cold, oral cobalt chloride and other complex factors can replicate the rat model of high-altitude polycythemia.

  Modeling method: SD rat, weighing 180-220g, male. The rats in the model group were kept in the "high altitude environment simulation cabin". There are three freezers in the cabin, which use decompression and hypoxia to simulate high temperature and low oxygen plateau environment. The temperature inside the cabin is lower than room temperature (9.0-11.0) °C. Close the cabin door and use the vacuum pump to reduce pressure. During the decompression process, the airflow in the air intake must be adjusted to keep the wind speed in the cab relatively high. Perform gradual intermittent decompression according to the altitude indicated by the "plateau barometer" in the cabin. The animal was decompressed for about 6 hours every day, and immediately after leaving the cabin, the animal was given a 15 mg/kg body weight of cobalt chloride aqueous solution. Sunday is a holiday, 40 consecutive days. Control rats eat freely at room temperature.

  Observation indicators and analysis:

  1, determine the hemoglobin content, hematocrit and red blood cell count;

  2, whole blood viscosity measurement;

  3. The influence of various model construction cycles on model construction. And observe the possible maintenance time under the environmental conditions of 2260 m;

  4. Pathological examination of lungs and kidneys.
