【Disease Animal Model】-Induced Liver Cancer Animal Model

[Modeling mechanism] The animal model of inducible liver cancer is a liver fiber that acts on experimental animals through chemical, physical and biological carcinogenic factors to induce the formation of animal liver cancer. The disease is a simulating periodic change. Currently, it is mainly induced by chemical carcinogens. Commonly used substances that cause liver cancer are diethylnitrosamine (DEN), dimethylaminoazobenzene (DBA), 2-acetylaminofluorene, 2-AAF, and o-aminoazotolue (OAT). ), Aflatoxin. ).

  [Modeling method]

DEN induction model: Sprague Dawley rats or Wistar rats are administered by forcibly orally 0.025% DEN aqueous solution at a dose of 10 mg/kg once a week. . A solution for animals to drink every day for 6 months of continuous feeding. Or, intraperitoneal injection of 0.5% DEN sterile aqueous solution (100 mg/kg). After 1 week, continue to provide 100 mg/L DEN sterile aqueous solution and drink for up to 16 weeks for free. Replace within 9-11 weeks. Used to disinfect water. DEN can also be used in combination with 2-AAF or Solt-Farber cancer induction methods. The rats were injected intraperitoneally with 200 mg/kg DEN solution, and after 2 weeks, most livers were given a 0.02% 2-AAF diet for 14 days. Resection was performed in the 3rd week, and normal feeding was resumed in the 4th week.

DBA-induced model: feed 0.06% DAB feed, control vitamin B2 content (≤1.5-2.0mg/kg), and feed continuously for 4-6 months. 2-AAF induction model: Sprague Dawley rats were fed 0.05% 2-AAF diet. Lasts 3-4 months.

OAT induction model: Apply 1% OAT solution (10 mg/ml) on the skin between the shoulder blades of the mouse every other day for 2-3 drops for 7-8 weeks. AF-induced model: The monthly diet of rat liver cancer is 0.001 to 0.015 ppm in 6 months. In addition, dimethyl sulfoxide (DMSO) can also be used to lyse AF, intraperitoneal injection of 400μg/kg AF once a day, stop the injection for 2 consecutive weeks, and administer 0.75 to 1.50 mg/kg AF once. Scale liver resection. Two weeks.

[Model Features] DEN is the most common chemical carcinogen, which induces a carcinogenic rate of 80% in rats. Liver cancer can occur within 6 months of the DBA method, and the incidence of cancer is 60%. Precancerous lesions will appear within 3 weeks after the induction of 2-AAF. Under the microscope, liver cells proliferate in oval cells with cancerous nodules of various sizes. After 3-4 months, 80%-90% of the animals are developing. .. Liver tumors. Nodules appeared 8 weeks after the administration of OAT smears, and liver cancer was induced in mice 7 months later, and the incidence of cancer was about 50%. Although the incidence of liver cancer induced by atrial fibrillation in rats is 80%, the disease course is longer and most tumors are diffuse nodules.

[Model Evaluation and Application] The induction model has a hidden beginning and a long induction cycle. Animal mortality during the induction process is relatively high, and there are great individual differences in the location of liver cancer, the number of lesions and the time. Currently, this model is mainly used to study the etiology, genetics and biology of liver cancer.
