【Disease animal model】-Traditional Chinese medicine-induced ventricular arrhythmia in rats

  (1) Copy method Experimental rats weighing 250-300g were anesthetized by intraperitoneal injection of 25% urethane and fixed in the supine position. According to the conventional method, needle electrodes were pierced into the rats' limbs and connected to the ECG machine and ECG oscilloscope, and recorded Lead Ⅱ ECG. First record the normal ECG before administration. Then intraperitoneal administration of Yinchen injection 14-21 mg/kg body weight (corresponding to 20-30 g crude drug), and observe the changes of the electrocardiogram. After the animals were given Yinchen, the sinus rhythm was significantly slowed down or disappeared, Pu's fiber self-discipline increased, ventricular premature, ventricular tachycardia and even torsade de pointes appeared. The arrhythmia appeared 2 to 10 minutes after administration. The preparation of Yinchen medicinal solution is to take Yinchen, add appropriate amount of water and decoc 2 times, each time for 10-15 minutes. Combine the two liquid medicines, filter with double gauze, and concentrate the filtrate to an appropriate amount.

  (2) Features of the model This model has the characteristics of rapid arrhythmia generation and long maintenance time, which is beneficial to observe the long-term therapeutic effect of drugs on arrhythmia.

  (3) Comparative medicine: The dry young stems and leaves of Asteraceae Artemisia genus A perennial herb. It has the effect of dehumidifying and relieving yellow. It has been clinically reported to promote choleretics, protect the liver, lower blood lipids, expand coronary arteries, lower blood pressure, and relieve fever. And antibacterial effect. However, when studying its effect on the heart of rats, it was found that it can cause arrhythmia in rats at a certain dose, and increase the dose can also cause ventricular fibrillation. It can be used in animal models of ventricular arrhythmia to study anti-ventricular arrhythmia drugs.
