【Animal Modeling】-an animal model of chronic gastric ulcer induced by duodenal reflux surgery

  (1) Breeding method Adult rats anesthetized with sodium pentobarbital were fasted for 24 hours without taking water, a midline incision was made in the supine position, the abdomen was opened, left and right anastomosis, and fasting for 3 days after the operation. 4 weeks after gastrointestinal anastomosis. Reopen the abdomen, cut the duodenum at the distal end of the bile duct opening, and then implant the two broken ends. Twelve weeks after the reflux surgery, the abdomen was anesthetized, the front wall of the front stomach was punctured, the gastric juice was drawn on an empty stomach, centrifuged, and the pH of the supernatant was measured. The probe electrode is inserted in the direction of the large and small curve to measure the blood flow of the two parts of the gastric mucosa. The blood is drawn from the inferior vena cava, centrifuged, and the serum is prepared. The serum gastrin content is analyzed. Collect the entire stomach. Make an incision along the big curve and observe the mucosal lesions. The anterior wall tissue of the vestibular stomach curve and the glands parallel to the branches was removed for histological morphological examination, and the thickness of the mucosa was measured.

  (2) Model characteristics: The pH value of the gastric juice in the model animal increases, the blood flow in the gastric mucosa decreases, and the serum gastrin level increases. Pathological examination revealed multiple round or oval ulcers in the lesser curvature of the gastric antrum, with clean edges, necrotic exudate at the bottom of the ulcer, and mucosal edema and thickening in the non-ulcer area. Area and fundus gland area. The area of the gastric antrum ulcer with small curvature was (7.4±2.9) mm2, and the mucosal thickness of the pylorus and fundus of the stomach increased by 29% and 35%, respectively.

  (3) Comparative medicine When the contents of the duodenum return to the stomach, it will cause inflammation and damage to the gastric mucosa, and the damaged gastric mucosa is easily damaged by acid and pepsin. According to the mechanism of the duodenal contents damaging the gastric mucosa, this experiment regurgitated all the duodenal contents into the stomach through surgery, thus creating a typical gastric antral ulcer model. It has the common parts and morphological characteristics of human gastric ulcer. Histology showed that the pathological changes of the ulcer and human gastric ulcer were very similar. Pathophysiology is more consistent with the natural development process of gastric ulcer formation.
