[Animal model]-Rabbit ear acne formation model

[Modeling mechanism] Acne is a combination of many factors, such as keratin growth of hair follicle gland epithelial cells, enhancement of cell adhesion, microbial infection, inflammatory damage of blood vessel walls, sebum and dandruff. The experimentally induced rabbit ear acne model, after ultrastructural analysis, its pathological mechanism is the proliferation of hair follicle keratinocytes, delayed shedding of keratinocytes, acne formation and hair follicle opening. And increase skin inflammation. It is partly similar to the formation mechanism of human acne.

[Model Features] Apply 50% oleic acid liquid paraffin to the 4 cm x 2 cm area of both ears of New Zealand rabbits once a day for 14 days. Histopathological sections are used to observe the expansion of the hair follicle opening, the size of corneal embolism, the degree of epidermal keratinization, and the degree of skin inflammation for qualitative and semi-quantitative judgments. In order to observe the efficacy of the drug, the inducer was applied to the induction area of the left ear for 1 hour at a time, and then the test drug was applied for 14 consecutive days, and the right ear applied substrate was used as a control.

[Evaluation and application of the model] The acne formation mechanism of this model does not include infection, inflammation and abnormal sebaceous gland secretion mechanisms. This is mainly epithelial hyperkeratosis and changes in keratin adhesion. However, according to the pathological indicators of optical and electron microscopy, the efficacy of the anti-acne drug retinoic acid is more consistent with the clinical acne formation and acne dissolution process.
