  [Operation steps] Add F344 or Wistar rats (body weight 70-150 g) and estrone (E1) to sterile castor oil, mix them thoroughly, and inject them into the abdominal cavity of experimental rats. Method 1: High-dose and short-term methods (10 mg each time, once a week). Method 2: Multiple low doses (2 mg each time, twice a week). A total of 30 to 60 days of injection.

  [Result analysis] There was almost no change in body weight, the weight of the whole brain did not change much, and the weight of the pituitary gland increased significantly, reaching about 34 mg (control: about 11 mg). The pathological analysis of the pituitary gland has the following three conditions: (1) the pituitary gland is not abnormal in shape, but the weight of the pituitary gland is significantly increased; (2) the pituitary gland is proliferatively changed, the arrangement structure still exists, but the cell density increases, and the cytoplasm swells. No nucleus atypia; (3) Tumor formation, the tumor of F344 mice has 70% tumor changes, the normal sinusoidal curve and the lack of space in the pituitary tissue, the destruction of the glandular sequence structure, the dense placement of tumor cells, and the cytoplasm is abundant Round polygon, showing nuclear atypicality, sometimes pituitary. When the drug was used for 45 days, the changes in brain glands were most obvious, and extending the study time did not increase the incidence of tumors (up to 60 days).
