【Animal Modeling】-Acute Visceral Inflammation and Pain Model

  1. Modeling materials Animals: healthy Wistar rats, male or female, weighing 240-270g, drugs: ether, 5% formaldehyde solution, saline, equipment: rectaloscope.

  2. Modeling method Ether gas is used to maintain the anesthesia of the rats. Rinse the rectal stool with warm normal saline at 37°C, and insert a special rectal endoscope through the anus. Under visual conditions, inject a mucosa of 0.1, 35 mm from the edge of the rectum. Ml 5% formaldehyde or 0.1 ml saline. Be careful not to penetrate the wall of the sigmoid colon during the injection. After the injection, it was slowly pulled out from the rectoscope to remove the anesthesia, and then the animal was placed in the observation cage.

Some fixed behavioral responses may reflect visceral pain: the rat licks the abdomen or genitals in a squatting position, slightly abducts the hind limbs, stretches the head to the lower abdomen, repeatedly licks or twists the lower abdomen or genitals, and licks the outside of the hind limbs, and the buttocks or hind limbs are quiet The ground moves and stops when disturbed; the abdomen contracts, the rat enters the hind limbs to stand, the abdomen contracts, bends backwards and leans to a high place, the center of gravity is forward, the buttocks are lifted from the ground, and the body trembles. The performance is relatively calm, even if you are annoyed by fear, you will not stop exercising. When stretching, the mouse’s front feet support the body, the back abdomen is flat on the ground, the hind limbs are stretched back, the whole body stretches, the muscles are tense and tremble, the performance is hurt, and the mouse hinders movement. The abdomen contracted and the mouse's entire body became flat. Support your body on the ground or forefoot, and flatten your back abdomen. Observed from the face, the inner side of the lower abdomen is flat. The limbs are close to each other. The ribs and abdomen are strongly contracted, showing a clear sense of depression, accompanied by tremors and physical fatigue, sometimes accompanied by stretching, pain and undisturbed by fear. Please stop.

  3. The behavioral response of modeling principle to visceral pain caused by inflammation caused by formaldehyde. 4. Changes after modeling Under anesthesia, the model rats were injected with formaldehyde under the mucosa of the sigmoid colon wall. After the anesthesia is restored, the behavioral response of visceral pain begins to appear, and the same behavioral response tends to be dynamic. It changes over time. In the experiment, the main symptoms observed were abdominal contraction (F), body extension (B), abdominal contraction (A) and scattered licking in the abdomen or peripheral areas of the body. (L), but the frequency of occurrence is very low. One hour later, L becomes the main behavioral response. Thirty minutes after the injection of formaldehyde into the pathological section of the model rat, a local ulcer was formed at the injection site, the superficial mucosal villous edema ruptured, the normal shape was lost, and the deeper part was observed. Stained inflammatory cells exuded on the mucosal surface, and there were local hematomas on the mucosa, and many deep-stained inflammatory cells infiltrated to form masses. The morphology of the muscularis mucosa and serosal layer is complete. The necrosis and dissolution of cells around the injection site resulted in a strong inflammatory response throughout the injection area.
