According to the different etiology and pathogenesis, Chinese medicine is divided into two types: Yang Huang and Yin Huang. [Modeling mechanism] The mechanism of the modeling method introduced in this article is based on the induction of liver injury in rats with 1-naphthyl isothiocyanate (ANIT), plus cold and humid environmental factors or combined with the traditional Chinese medicine rhubarb and Based on cold and damp environmental factors, an animal model of yin and jaundice in TCM was established.
【Modeling method】Experimental animal: Wistar rat. The experimental animals were intragastrically administered with 2ml 100g of 0.9% normal saline every day on the 1st to 21st day, and on the 22nd day with 0.5% ANIT once (dissolve ANIT in salad oil), and put it in the cold and wet model box (Temperature in the moulding box is 10~12℃, relative humidity>95%). Or put the test animals in the cold and wet modeling box (the temperature in the modeling box is 10~12℃, the relative humidity is >95%), and use 100% rhubarb decoction (about 1g per ml of crude drug) 2ml every morning /100g gavage. On the 22nd day, 0.5% ANIT was given by gavage once. Observation indicators: model animal body weight, rectal temperature, general biological status, β-glucuronidase content, glucuronidase activity, liver function changes, liver histopathological changes and humoral immunity levels.
【Features of the model】Observation of the general state of the rats during the modeling stage. From the 3rd day onwards, the model rats have squinted eyes, reduced activity, huddled up in groups, hid in groups, bowed back, withered, loose coat, and moistened. Obscure, loose stools, dirty perianal, weight loss, and rectal temperature. At 48 hours, the sclera of the jaundice model rat was yellowish, the skin on the back of the ear was slightly yellow, the tail was yellow, and the urine was dark yellow.
[Model evaluation and application] This animal model conforms to the characteristics of jaundice, that is, the modeled rats showed increased β-glucuronidase content, decreased glucuronidase activity and D-xylose content, and liver histopathological changes were consistent with The pathological changes of intrahepatic cholestasis and jaundice are in line with the main characteristics of ANIT gavage modeling method; at the same time, the model rats have obvious main symptoms of cold-dampness syndrome and changes in objective indicators. It conforms to the traditional Chinese medicine theories of etiology and pathogenesis, and the method is reliable and reproducible, similar to human jaundice. It can be applied to the study of traditional Chinese medicine etiology and pathogenesis theory, the mechanism of anti-jaundice Chinese medicine and the research and development of anti-jaundice medicine.
