Diabetes belongs to the category of "wasting-thirst" in Chinese medicine. The pathogenesis of Chinese medicine mainly includes yin deficiency and fire prosperity, lung and kidney yin deficiency, qi and yin deficiency, and kidney yang deficiency. At present, the widely used western medicine model of type 2 diabetes is streptozotocin (STZ) induction method. The most important thing in the experimental research of traditional Chinese medicine is the establishment of various "syndrome" animal models.
[Modeling mechanism] Deficiency of Qi and Yin is the most common type of diabetes. That is, after STZ causes diabetes (blood sugar>16.7mmol/L), the traditional Chinese medicine Qingpi, Citrus aurantium, aconite and other traditional Chinese medicines are used to make a model of deficiency of both Qi and Yin.
[Modeling method] Experimental animals: Sprague Dawley rats. The experimental rats were fasted with water for 12 hours, and STZ 30mg/kg was injected into the tail vein. On the 5th day, if the random blood glucose of the orbital venous plexus is >16.7mmol/L, the model of type 2 diabetes is successfully established. After taking the traditional Chinese medicine Qingpi and Citrus 18g each, 15g aconite, 13g/kg body weight, which is equivalent to 20 times the amount of human 1 time a day. Observe the general state of the rats after the rats are calm at 8-10 am and 24 pm every day. The weight is recorded every 2 weeks. The rats were sacrificed 4 weeks later, and the objective indicators were checked. Observation indicators: general biological status, changes in serum insulin, thromboxane B2, 6-ketoprostaglandin F1α levels.
[Characteristics of the model] After the model was created, the rats showed symptoms such as laziness, fat tongue, and less fluid. Serum insulin, thromboxane B2 and 6-ketoprostaglandin F1α levels changed. There were significant differences in insulin and thromboxane B2 between the model group and the diabetic control group.
[Model Evaluation and Application] This model animal has symptoms such as laziness, fat tongue, and less fluid, and the symptoms are alleviated after Zhonghui Tangmaikang is treated with counter-syndrome, which proves that the syndrome is consistent with the deficiency of both Qi and Yin. It can be applied to the theoretical research of the medicinal properties of Chinese medicine and the development of Chinese medicine preparations for diabetes.
