Objective: To optimize the dose of histamine plus 4-aminopyridine (4-AP) combination model by using the Starpoint design effect plane method to establish a new type of guinea pig pruritus animal model.
Method: Use Starpoint Design to arrange experiments. Each group injected 0.5 mL of different pruritus under the skin of the bald head. We recorded the number of scratches and scratches of the guinea pig in 30 minutes. And through the comprehensive scoring method to screen out the best itching formula. The guinea pig model is evaluated by observing the behavioral changes of the guinea pig and measuring the histamine and interleukin 6 (IL-6) levels in the blood.
Results: In the behavioral experiment, the number of scratches in the combination group within 30 minutes was significantly higher than that of the histamine group and 4-AP group (P\u003c0.01), and the scratch latency was significantly reduced. Compared with the control group, the serum histamine concentration of morphine in the combination group and histamine group was significantly increased (P\u003c0.05 or P\u003c0.01), but the increase in the combination group was higher than that of the control group. In the histamine group, it is very low. (P\u003c0.05); The serum IL-6 concentration of histamine, 4-AP and the combination group was significantly higher than that of the control group (P\u003c0.01 or P\u003c0.05). The group was significantly higher than the histamine group and 4-AP group. Pathological examination showed that the proliferation of inflammatory cells in each model group was different, and the response of the combined group was more obvious.
Conclusion: The formula (joint group) model optimized for this experiment is easy to implement and has good repeatability.
