  Objective: To explore the factors that influence the establishment of the tumor C57BL/6J mouse melanoma lung metastasis model, such as the method of tumor inoculation, the number of inoculated cells, and the cycle of tumor occurrence.

  Method: Culture mouse melanoma cells B16F10 in vitro; 1) Male mice aged 18 to 6 to 8 weeks were randomly divided into 3 groups, and 6 mice in each group were injected into the tail vein. After 2 weeks, 100 μL (3×106 cells) was injected intraperitoneally and subcutaneously to suspend B16F10 cells in each mouse. The mice were dissected and the growth and metastasis of melanoma were observed; 2) Same as above, divided into 3 groups and Inject 3×106 cells, 1×106 cells, 3×105, 2 weeks later, the cells passing through the tail vein were dissected to observe the growth and metastasis of melanoma; 3) Divide into three groups as described above and inject into the tail vein 1×106 cells, dissecting mice within 1 week, observe the growth and metastasis of melanoma within 2 and 3 weeks?

  Results: 1) After tail vein injection of mouse melanoma cells, the success rate of mouse lung metastasis was 100%, but neither intraperitoneal nor subcutaneous injection caused lung metastasis; 2) Inoculation of melanoma into mice if the number of cells If the number of melanoma cells transferred to the lung is 1 x 106, the number of melanoma cells transferred to the lung is moderate; if the number of cells infused is 3 x 106, the number of melanoma cells transferred to the lung is too much; When the number is 3×105, the number of melanoma cells transferred to the lung is very small; 3) 1×106 mouse melanoma cells are injected into the tail vein. After feeding for 2 weeks, we were able to observe obvious lung metastasis of melanoma cells without letting the mice die. After three weeks of feeding, too many melanoma cells transferred to the lungs, killing more than half of the mice. Is the number of melanoma cells transferred to the lungs very small within a week?

  Conclusion: The melanoma cells grown by tail vein injection of 1 x 106 mice for 2 weeks are the recommended method for constructing melanoma lungs in C57BL/6J mice.
