【Animal Modeling】- Rat Sepsis Model

Objective: How to study the changes in blood coagulation function of septic rats caused by pus puncture (CLP)?

Method: Use CLP to induce sepsis in SD rats, and detect the correlation at 8-16 hours and 48 hours after the operation. Are the coagulation function index, HE lung, kidney, liver, and spleen stained to observe histopathological changes?

Result: Is the 12-day survival rate of CLP-induced septicemia rats 30%, showing acute attacks and high mortality? In the acute phase of development, the APTT time of CLP rats was extended by 8 hours (P\u003c0.05), the PT time was extended by 16 hours (P\u003c0.05), and the XII activity and the external coagulation pathway of the endogenous Sex. There is a coagulation pathway. Transient suppression? Will TT be extended within 48 hours (P\u003c0.01)? The FIB content gradually increases from 16 hours (P\u003c0.001)? The number of PLT among other important indicators related to coagulation and anticoagulation function gradually decreased from 8 hours (P\u003c0.01); vWF: The number of Ag gradually increased from 8 hours (P\u003c0.01.u003c0.001) ). ; D? The content of D gradually increased from 16 hours (P\u003c0.05); PS: The content of Ag did not decrease significantly until 48 hours (P\u003c 0.001); Is there a significant change in the content of III in AT? Pathological examination showed different levels of damage to lung, kidney, liver and spleen tissues, but did not show any obvious venous thrombosis and bleeding characteristics.

Conclusion: The rat sepsis model has acute coagulation dysfunction, but is there any histopathological change caused by coagulation function?
