【Animal Modeling】-Improve the inflammatory model of Mycoplasma pneumoniae in mice

  Objective: To explore the therapeutic effect of Qingfei Touxue Decoction on mice with Mycoplasma pneumoniae and its mechanism.

  Method: 40 male SPF BALB/c mice were randomly divided into control group, model group, Qingfei Touxue Decoction low-dose group, middle-dose group and high-dose group, each with 8 mice for each group. A mouse model of Mycoplasma pneumoniae pneumonia was established by intranasal injection of Mycoplasma pneumoniae (MP). The mice in the model group were given physiological saline. The Qingfei Touxie Decoction suspension was administered 800 mg/kg body weight daily. After 2 weeks of treatment, the lung tissue index of the mice was calculated. HE staining is used to detect pathological changes in lung tissue. TUNEL is used to detect apoptosis in mouse lungs. ELISA was used to detect IL-1β and TNF-α. , And IL in the alveolar lavage fluid of each mouse. -6, IL-18 content; qPCR to detect the expression of NLRP3, MyD88 and NF-κB mRNA in lung tissue; Western blotting to detect the expression of NLRP3, MyD88 and p-NF-κB protein in lung tissue.

  Results: Qingfei Touxie Decoction can significantly reduce the lung index of MPP mice, reduce lung pathological damage and cell apoptosis rate (P\u003c0.05); Qingfei Touxie Decoction can improve the effect of alveolar lavage fluid, and also Can significantly reduce α and β IL-1β and TNF. IL-6,, IL-18 levels (P\u003c0.05); and NLRP3, MyD88 and NF-κB mRNA and protein expression decreased (P\u003c0.05).

  Conclusion: Qingfei Touxue Decoction can improve lung injury in mice with mycoplasma pneumonia, and its mechanism of action may be related to the inhibition of NLRP3 inflammasome and NF-κB signaling pathway.
