【Animal Modeling】-Heart Failure Rat Model of Heart and Kidney Yang Deficiency

  Objective: To study the autophagy mechanism of Bugu Decoction on myocardial cells in rats with heart and kidney deficiency and yin and yang deficiency.

  Method: Surgical removal of the thyroid gland and intraperitoneal injection of adriamycin can establish a rat model of heart and kidney yin and yang deficiency, and inject it into the stomach for 10 days. Detect and record the natural state of each group of rats, and use ELISA to detect serum triiodothyroxine (T3), thyroxine (T4), thyroid-stimulating hormone (TSH), myocardial tissue BNP, LA, and CaN content. Westernblot studied the expression of LC3, ATG5 and P62 proteins. The expression of LC3 and P62 in rat myocardial tissue was determined by immunohistochemical method. HE staining to observe the pathological changes of myocardial tissue.

  Result: Zhenwu Decoction can reduce the content of BNP in myocardial tissue, increase the content of LA in myocardial tissue, reduce the content of CaN, increase the content of LC3, ATG5, BECLIN1 in myocardial tissue, and reduce the content of P62.

  Conclusion: Genbu Decoction can improve heart failure caused by insufficient heart and kidney yin and yang, and improve heart energy metabolism. In the early stages of heart failure, autophagy can be regulated to protect damaged cardiomyocytes.
