[Animal Modeling]-Rats with Ocular Ischemic Syndrome

  Objective: To establish a rat model of ocular ischemia syndrome and observe the morphology of the retina after ischemia and the difference in the expression of RhoA and ROCK-2 in the rat retina.

  Method: 20 Rattus norvegicus were randomly divided into OIS model group and sham operation group. The rats in the OIS model group were modeled by complete ligation of bilateral common carotid arteries (BCCAO). The rats in the sham operation group were released only without ligation. The number of retinal ganglion cells and the thickness of each layer of the retina of the two groups of rats were observed after 3 months of modeling by hematoxylin-eosin (HE) staining. RhoA and ROCK were analyzed using immunohistochemistry and Western blot. The expression level of rat 2 in the retina of the two groups.

  Result: 9 rats in the model group were successfully modeled. Compared with the sham operation group, the number of retinal ganglion cells in the model group was significantly reduced (P0.05). Compared with the sham operation group, the expression levels of RhoA and ROCK-2 protein in the retina of the model group increased significantly (the former is P\u003c0.01, the latter is P\u003c0.05).

  Conclusion: The expression of RhoA and ROCK-2 increased significantly in the OIS model group, which may provide new ideas for the prevention and treatment of the disease.
