[Animal Modeling]-Coronary Heart Disease Model

  Objective: To establish a rat model of type 2 diabetes with coronary heart disease.

  Method: A Zucker diabetic obese rat (Zuckerdiabeticfatty, ZDF) model of type 2 diabetes with coronary heart disease was prepared by feeding a high-fat diet and continuously injecting isoproterenol hydrochloride (ISO). The model was evaluated by serum creatine kinase (CK), serum creatine kinase isoenzyme (CK-MB), ECG ST and cardiac tissue morphology.

  Results: The expression of CK in the non-DM model group and the DM-CHD group gradually increased with the increase in the frequency of administration. The expression of CK-MB first increased and then decreased, and myocardial damage was formed in rats. The ST segment of Reed II of the model rat ECG showed deviation, indicating that myocardial injury was formed. The results of pathological section of myocardium of rats in the injection model group showed that the myocardial cells of the blank control group were normal. The area of myocardial necrosis in ZDF rats was the cross-sectional area of the myocardium after continuous injection, reaching 1/2 ISO for 5 days4

  Conclusion: CK, CK-MB content changes, ECG ST wave changes and myocardial pathological sections indicate that the model is successful. ISO may cause some myocardial damage. Continuous infusion of ISO can be used to establish a coronary heart disease model. In this article, we have established a simple method to construct a rat model of diabetes and coronary heart disease with a high-fat diet and continuous injection of ISO.
