【Animal Modeling】-Using orthogonal experiments to optimize the establishment of a conditioned fear memory model

Purpose: Use orthogonal experiments to optimize the best experimental parameters and find the best experimental conditions to establish a conditioned fear memory model in rats?

Method: Use Paprov’s conditioned fear principle to establish a rat conditioned fear memory model. Three elements are used. The horizontal orthogonal test design takes the rat's fear reaction time 24 hours after the establishment of the 3 models as an indicator, observes the development of fear memory under various parameter conditions, and determines the best test conditions? Establish a conditioned fear memory model with the best experimental conditions. , And the fear memory retention test after 24 hours? 1 week? Two weeks? 4 weeks and 8 weeks?

Result: The intuitive analysis results show that the degree of influence of each factor is in the order of sound intensity = number of cycles → electric shock. The results of variance analysis show that the number of cycles is the result of the experiment, indicating that its influence is greater. (P0.05)? Determine the best experimental conditions as follows: sound 75 dB, electric shock 0.8 mA, 15 cycles? In the case of the model, it was established for 24 hours and 1 week. Compared with the control group, the fear memory of the rats was well maintained. Are there significant differences between groups (P0.05)?

Conclusion: This experiment revealed the primary and secondary factors of conditioned fear in rats. It optimizes the memory model, experimental conditions, and provides model standardization, standardization and follow-up reference.
