[Animal Modeling]-Sterile APPswe/PS1ΔE9 double transgenic mouse model

Objective: To establish a sterile APPswe /PS1ΔE9 (PAP) double transgenic mouse model by cesarean section, and to initially observe the plaque deposition in the animal's brain, in order to study the relationship between the intestinal flora and Alzheimer's disease Provide new ideas.

Method: select positive PAP male heterozygous mice and deliver C57 wild-type female mice for 1:2 mating? Can pregnant female mice undergo super-cesarean section clean bench breast replacement with sterile ICR mice? In the brains of 9-month-old PAP mice, sterilization tests were performed monthly after surgery; PCR was used to detect the genotype of PAP offspring obtained by caesarean section; and immunohistochemistry was used to quantitatively detect changes in plaque. : 12 cases were delivered by caesarean section, 66 pups were collected. Survival rates of caesarean section and weaning were 95.45% (63/66) and 95.24% respectively (after purification, sterility was tested in accordance with national standards. Are all states certified? The results of immunohistochemistry showed that 9-month-olds had no PAP mice have fewer hippocampal plaques.

Conclusion: Cesarean section technology can clear the bacterial flora of PAP mice. Can it reduce the plaque in the brains of 9-month-old infertile PAP mice?
