【Animal Modeling】-Allergic Asthma Model

OBJECTIVE: To explore the method of establishing an asthma model in which antibiotics cause airway flora imbalance, and gradually discover the mechanism of traditional Chinese medicine to treat diseases by regulating the flora, which has traditional significance for airway flora and asthma.

Method: 40 SPF male BALB/c mice were randomly divided into 4 groups: normal control group, asthma model group, nasal antibiotic asthma model group, and intraperitoneal injection of antibiotic asthma room dust. ), cefoperazone and sulbactam sodium (CFP) were administered by nasal drip or intraperitoneal injection, respectively, and the effect of antibiotics on HDM-induced asthma was observed. Twenty-four days later, blood was drawn from the can of the mice, and there was very little blood in each group. Indicators of mouse asthma: counting eosinophils (EOS) in the blood, testing serum, mouse alveolar lavage fluid (BALF) and TH2 cytokine IL-5 IgE levels, and lung tissue in mouse HE lung tissue Homogenized IL-13 level? Mason? Have you stained with PAS and observed histopathological changes?

Result: EOS in the serum of the mouse asthma model group? IgE? The levels of BALF and IL- in lung homogenate 5 and IL-13 were significantly higher than those in the blank group (P\u003c0.05), and antibiotics administered in the nasal infusion group were significantly higher than those in the asthma model group (P (u003c0.05). Histopathological observations showed that antibiotics were used for nasal infusion to establish a model. Is there inflammation in the group? Compared with the model group, the intraperitoneal antibiotic group had more serious extracellular matrix deposition and mucus secretion, and related asthma indicators There is no obvious change.

Conclusion: The antibiotics cefoperazone and sulbactam sodium are used for intranasal administration. Mice can aggravate HDM-induced asthma in mice, indicating that the administration of antibiotics to the airway can cause airway obstruction. This model suggests that if GF mice are not reproduced, it can be used as a model for studying the airway flora and asthma regulation of traditional Chinese medicine.
