How to build a learning and memory impairment model? -Animal modeling

  Objective: To study an effective method to establish a mouse model of learning and memory impairment through light protection and Morris water maze experiments.

  Method: 20 minutes before the electric shock avoidance training, 1 mg/kg Dongpol base was injected into the abdominal cavity of the mice. After the electric shock training, the mice were injected subcutaneously with 90 mg/kg sodium nitrite. A model of 0.1 mL/10 g of ethanol, memory acquisition, memory integration, and memory dysplasia was established 15 minutes before the next electric shock test. Each group of mice has a 6-hour blackout latency, error time and detection time. To evaluate the sensitivity and optimal test time of each of the three memory impairment model methods, the measurements were carried out at 24 hours, 30 hours and 48 hours after the electric shock training. The mice were injected intraperitoneally with 1 mg/kg Dongpol base and 0.1 mL/10 g 35% ethanol, which caused learning and memory impairment. Use Morris water maze positioning navigation test and space exploration test to test the sense of spatial position in experimental animals. Establish the direction (spatial direction) of the evaluation method for the mouse model of spatial learning and memory impairment. The results of the dark place avoidance experiment showed that the memory impairment model mice induced by Eastpol alkali significantly reduced the latency of avoiding the dark place at 6 hours and 24 hours after the electric shock training, and the number of errors was also high. Significant increase (P0.05).

  Conclusion: In the dark experiment, we should use Dongpol base and sodium nitrite to establish a model of memory acquisition impairment and memory integration in mice, and test the corresponding indicators 24 and 30 hours after modeling. After administering ethanol to the mice 24 hours after the electric shock training and 15 minutes before the next test, the ethanol-induced memory/aplastic disorder model was tested. The metric is to avoid latency in the dark, the number of errors, and the number of probes. The Morris water maze experiment showed that Eastpol base may impair the spatial learning and memory of mice, and it can be used as a medicine to establish a model of spatial learning and memory impairment in mice.
