What is the method to inhibit the formation of blood vessels in liver cancer model rats? -Animal modeling

  Objective: To investigate the inhibitory effect of Zhenqi Fuzheng Granule on angiogenesis in liver cancer model rats and its possible mechanism.

  Methods: Wistar rats were divided into control group (control group), model group (prepared by low-dose diethylnitrosamine (DEN) induction method), Zhenqi Fuzheng granule group (2.62g/kg) and si-miR -200c group (20 o'clock in the afternoon). Zhenqi Fuzheng Granules + si-miR-200c group, 12 mice in each group, each group was treated for 8 weeks. HE staining to observe the pathological changes of rat liver tissue; enzyme-linked immunoassay to detect serum vascular endothelial growth factor (VEGF), alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels; real-time fluorescence Quantitative PCR method to detect the expression of miR-200c in liver tissue; Western blot method to detect the expression of zinc finger protein (ZEb-2) and VEGF protein in liver tissue; ZEb-2, VEGF and immune tissues with positive expression of microvessel density (MVD) chemical method.

  Result: Compared with the control group, the model group has higher levels of VEGF, ALT and AST, and more deformed and necrotic cells in the liver tissue. The expression of miR-200c in liver tissue decreased, while the expression of ZEB-2 and VEGF protein increased. The positive expression of ZEb-2, VEGF and MVD increased (P\u003c0.05). Compared with the model group, the Zhenqi Fuzheng Granule group had lower levels of VEGF, ALT and AST, smaller cell deformation, higher differentiation of cancer cells, and increased expression of miR-200c. , ZEb-2 and VEGF protein expression decreased, and ZEb-2 expression decreased. In the si-miR-200c group, the positive expression of VEGF and MVD decreased; the levels of VEGF, ALT and AST decreased, and the expression of miR-200c in liver tissue decreased, while the expression of ZEB-2 and VEGF protein increased. 2. The positive expression of VEGF and MVD increased (P\u003c0.05). Zhenqi Fuzheng Granule + si-miR-200c group VEGF, ALT, AST levels are lower than si-miR-200c group, ZEB-2, VEGF protein, ZEB-2, VEGF, MVD positive expression rate are higher than si-miR-200c group. In the low miR-200c group, the expression of miR-200c was higher than that of the si-miR-200c group (P\u003c0.05).

  Conclusion: Zhenqi Fuzheng granules can inhibit angioplasty in liver cancer model rats and reduce its development. This mechanism may be related to the up-regulation of miR-200c expression and negatively regulate the expression of ZEb. -2 protein.
