[Animal model]-Tissue thromboplastin-induced rabbit acute DIC model

  (1) Method of replication: Experimental rabbits weighing 2~3kg, give a dose of tissue thromboplastin 20mg/kg body weight into the rabbit ear vein, and determine before and immediately after the injection of tissue thromboplastin at 1, 3, 6, 12 and 24h Whole blood clotting time (test tube method), partial thromboplastin time, platelet count, plasma fibrinogen content, serum fibrin (proto) lysate, euglobulin lysis time and other indicators; model animals after tissue thromboplastin injection Pathological examinations were performed at 1, 3, 6, 8, and 12 hours. Heart, lung, liver, spleen, kidney and adrenal glands were taken, and stained with HE and Mallory cellulose.

  (2) Features of the model This model has the characteristics of rapid response and violent response, and the surviving animals recover quickly. The main pathological change is extensive microthrombosis in the lungs. The changes immediately after injection are the most significant, which takes a short time and has obvious effects.

  (3) Comparing the DIC model caused by medical injection of tissue thromboplastin, it showed that the clotting time and partial thromboplastin time were significantly prolonged, but the latter was more significant and the duration was longer; fibrinogen and platelets decreased rapidly, while serum Fibrin (proto) cleavage products increase rapidly. Fibrinogen and fibrin (proto) cleavage products showed negative correlation changes, similar to clinical DIC changes.
