How to create a mouse ZP2 antibody? -【Animal Modeling】

  Objective: To prepare anti-mouse ZP2 antibody and study the immune activity of antiserum.

  Methods: The Rosetta host strain was used to induce the expression of mZP2 recombinant protein, the recombinant ZP2 protein was separated by electrophoresis, and the gel was cut to prepare immune antigen; the emulsified antigen was used to actively immunize New Zealand male rabbits to prepare antiserum; the antibody response level was determined by ELISA; the antiserum was determined by immunohistochemistry Specificity: The effect of antiserum on the sperm of mice after the egg-binding acrosome reaction was detected by the sperm-egg binding experiment.

  Result: ELISA results indicate that immunization of New Zealand male rabbits with recombinant protein induces antiserum. The immunohistochemical experiment proved that the antiserum had a specific reaction with the zona pellucida of the mouse. The sperm-egg binding experiment showed that the anti-ZP2 antibody could inhibit the sperm-egg binding after acrosome reaction.

  Conclusion: The recombinant mZP2 protein induces the production of anti-ZP2 antibodies, which can inhibit the binding of sperm and eggs after acrosome reaction.
