How to study the virulence of neonatal mice of different clinical strains of EV71? -【Animal Modeling】

Objective: To study the toxicity of various EV71 strains to newborn mice.

Method: Inoculate clinically mild, severe and dead EV71 strains, 1-day-old BALB/c infant mice, observe the symptoms of the mice for 14 days, and the mice for 1, 3, 5, 7 and 9 days. Organize RT after infection? The viral load was measured by QPCR, and the lesions were observed by HE staining.

Result: There was no significant difference in the severity of symptoms between the severe and mild strains (P = 0 693), and both groups were heavier than the dead strain (P = 0 6). The mild, severe, and mortality groups were 77-2%, 81-7%, and 97-8%, respectively, and the difference was statistically significant (000, P = 0≤000). P = 0.0010, P = 0 001, P = 0.0004); the pulmonary hemorrhage in the mild strain group was the most severe, and there were no significant differences in the pathological characteristics of the brain, muscle, spleen or intestines; among the tissues in the mild strain group The virus has the lowest proportion of the highest number of people and the lowest number of deaths.

Conclusion: The clinically mild virus strain is the most serious in lactating mice, and the number of deaths is very small. It is inconsistent with human clinical results and may be related to viral genes, host and other factors.
