【Animal Modeling】-What are the pathological mechanisms of the asthma mouse model under the action of paeoniflorin?

  Objective: To observe the effect of medicinal glycosides on airway inflammation chemokines and their receptors in a mouse asthma model.

  Method: Use OVA BMIN (OVA) to sensitize and challenge to establish a mouse asthma model. Serum IL-6, TNF-α levels and bronchoalveolar lavage fluid (bronchial alveolar lavage fluid, BALF) egg protein specific IgE (OVA)-ELISA method for detecting IgE) and chemokine CCL19, CCL21 levels; lung tissue Chemokine receptor-PCR method in RT detects human CCR7 mRNA expression; Western blot method detects lung tissue CCR7 and nuclear factor-κB (nuclearfactor-κB, NF-κB) protein expression.

  Results: The serum IL-6 and TNF-α levels of the mice in the mucorin intervention group were significantly reduced; BALF OVA-IgE, CCL19 and CCL21 levels were significantly reduced; CCR7 mRNA, CCR7 and NF-κB in the lung tissue were significantly reduced cut back.

  Conclusion: Pe medicinal glycosides have a significant inhibitory effect on airway inflammation chemokines CCL19/CCL21 and its receptor CCR7 in asthma model mice.
