【Animal Modeling】-Why establish a metabolic profile model of liver tissue in rats with chronic heart failure?

Objective: To establish a model of the metabolic characteristics of liver tissue in rats with chronic heart failure, to study the changes in liver metabolism of chronic heart failure from the perspective of metabolism, to explore valuable metabolites in the molecular mechanisms of heart failure and disease, and to treat them.

Method: 20 male Wistar rats, the heart failure group underwent abdominal aortic stenosis surgery, and the control group underwent sham surgery, kept for 24 weeks. Use the metabolomics research platform to analyze the metabolic characteristics of rat liver tissue, establish an orthogonal partial least squares discriminant model and a principal component analysis model of heart failure rat liver tissue, and finally combine the data processing with SPSS 19.0 software to obtain metabolism The nature of things.

Result: We successfully established a principal component analysis model and an orthogonal partial least squares discriminant model. At the same time, we screened and identified 10 different metabolites, including lysophosphatidyl bile, which are different between the heart failure group and the control group. Alkali, lysophosphatidylethanolamine, oleic acid, glycocholic acid, dehydroepiandrosterone sulfate and other substances.

Conclusion: The established metabolic model is in line with the liver metabolism disorder in rats with chronic heart failure. The selected characteristic metabolites can provide reference and support for the pathophysiological molecular mechanism of the disease and the diagnosis and treatment of the disease.
