【Animal Modeling】-How to establish a rat model of diarrhea-type irritable bowel syndrome?

  Purpose: To observe changes in rats’ symptoms, histopathology, visceral hypersensitivity, mast cell activation, autophagy, beclin-1 and claudin-2 and other related indicators for capsaicin gavage. Symptoms of diarrheal irritable bowel syndrome (D-IBS)) evaluate the combined restraint stress, and conduct new research on the model.

  Method: Forty male SD rats of clean grade were divided into normal group, group I, group II and group III according to their body weight, with 10 rats in each group. In the model group, the restraint tail clamp stress was combined with capsaicin (0.125% in group I, 0.250% in group II, and 0.500% in group III), and a D-IBS model was established by intragastric administration. Inject the same dose of saline into the stomach twice in a row. Powerlab was used to detect the contraction of the rat's abdominal wall and the number of dorsal arches, mast cell staining (Fuxin-Orange G method) was used to detect the activation of mast cells, and the pathology and autonomy of the rat’s colon were examined with light microscope and electron microscope. Immunohistochemistry SABC was used to detect the changes of phage to detect the changes in the expression of beclin-1 and claudin-2 in the colon mucosa of rats.

  Result: All rats in the model III group died, the number of feces in the model II group increased, and the visceral sensitivity threshold decreased. Compared with the normal group and the model group I, the difference was statistically significant (P\u003c0.05). ); each group of rats had normal colonic mucosa, mucosal epithelial villi and mucosa, submucosal vasodilatation and diffuse inflammation infiltration were unchanged; mucosal matrix or submucosal tissues showed increased expression of mast cells, until dark purple spots, looking like It looks like a round purple red; the autophagy expression of Beclin-1 and claudin-2 in the colonic small intestine epithelial cells of the model II group was significantly increased compared with the normal and model I groups, and the difference was statistically significant (P\u003c0. 05).

  Conclusion: The rat D-IBS model established by the combination of restraint stress and 0.25% capsaicin forced feeding showed increased diarrhea, visceral hypersensitivity, increased mast cell expression, increased intestinal epithelial cell autophagy and intestinal tract, showing mucosa The barrier is broken.
