【Animal Modeling】-What is an endometriosis model?

Objective: To explore the effects of different doses of estrogen on the treatment and recurrence of endometriosis?

Methods: Divide 51 endometriosis model rats into 5 groups: control group (n=10), a total of 1 mg/kg saline injection once subcutaneously; research group: anti-additive group I (low-dose estrogen group = 10)? Anti-addition group II (medium-dose estrogen group) n=10)? Anti-addition group III (high-dose estrogen group n = 10)? GnRH-a group (n = 11)? The test group was given a subcutaneous injection of 1 mg/kg leuprolide, I, II and III. Two weeks after administration, estradiol valerate (25μg/kg~50μg/kg~100μg/kg) was given, and each test group in GnRH-a group was given normal saline once a day. The volume of abnormal internal lesions was measured at 4 and 7 weeks after administration, and internal non-lesional tissues were collected, and internal TNF was detected by immunohistochemical method. -α? StAR expression, blood sampling from the intraorbital venous plexus, and enzyme-linked immunosorbent assay to detect serum CA125 levels

Result: There was no significant difference in the number of endometriosis before treatment. Rat ectopic lesions in each group ((P→0.05) The study group was significantly smaller than the control group at the 4th week, and the difference was significant (P0.05). At the 4th week, the TNF-α expression α? The test group StAR was significantly lower In the control group, significantly (P0.05)? In the 4th week, the test group serum CA125 was lower than the control group, the difference was significant (P u003c0.05)? In the 7th week, the anti-addition group II?III was higher than the anti-addition group. Addition group 1, GnRH-a group serum CA125 level is higher, the difference is significant (Pu003c0.05)?u003c 0.05)?

Conclusion: GnRH-a reversely adding low-dose estrogen has a positive effect on the rat endometriosis model. When the middle and high-dose estrogen is added reversely, TNF-α is added to the hormone.
