【Animal Modeling】-How to establish a rat model of chronic inflammatory pain?

  OBJECTIVE: To dynamically observe the changes of pain mood in rats with chronic inflammatory pain induced by Freund's complete adjuvant (CFA), and to explore the time lag induced by anxiety and depression in rats with chronic inflammatory pain.

  Method: All experimental rats were randomly divided into a blank control group and a model control group. The model control group was injected with CFA at the sole of the foot to establish a chronic inflammatory pain model. Dynamic observation of the changes in mechanical withdrawal threshold (PWT) of rats in each group before and after modeling for 1, 3, 7, 18, and 26 days; 28-day open field test (OF), 29-day elevated O maze test (EZM) observation)) , New Food Control Test (NSF) on Day 30, Marble Burial Test (MBT) for simulated 12, 20, and 31 days, and Sugar Preference Test (SPT) for simulated 17, 25, and 35 days.

  Results: 1 day after modeling, the PWT of the model control group was significantly reduced (P0.05); only 31 days after MBT modeling, the number of marbles embedded in the model control group was more significant than that of blanks. The control group increased (P0.05).

  Conclusion: The rats in the CFA-induced chronic inflammatory pain model can show various animal behavior changes related to anxiety-like emotions after 4 weeks, but they will develop depression-like emotions within 5 weeks, and no correlation is observed. Changes in animal behavior.
