【Animal Modeling】-How to establish a MuSK myasthenia gravis mouse model?

  Objective: To establish a mouse model of myasthenia gravis with positive muscle-specific receptor tyrosine kinase antibody (MuSK-Ab).

  Method: 6-8 weeks old eagle-like C57BL/6 mice (40 mice). Random grouping and modeling are used for weekly reproduction. On the first day of modeling, immune antigens (including 50 μL, 12.5 μg mouse peptide and CFA complete Freund's adjuvant for each site) were injected into 4 sites (2 hind legs and 2 shoulders). Modeling reinjects the immune antigen into the above four positions (two hind legs and two shoulders), and records the daily morbidity from the 70th day.

  Result: The success rate of the model is 50. The score is 1 mouse 1 point, 3 mice 2 points, 5 mice 3 points, and 1 mouse 4 points.

  Conclusion: The creation of the MuSK-Ab mouse model is feasible and will help the detailed study of myasthenia gravis.
