【Animal Modeling】-Analysis of Biological Parameters and Cardiac Physiological Function of Slit2 Gene Overexpression Mice

  Objective: To measure and evaluate the basic biological parameters, echocardiographic morphology and transcriptome sequence of human Slit2 gene overexpressing mice (Slit2 Tg mice), and to provide reference materials for the application of mice in biomedical research.

  Method: Slit2 was bred from inbred lines? Observe and compare the reproductive status of Tg and C57BL/6J mice, use PCR technology for genotyping, regularly draw blood for blood biochemical examination, protein expression and pathological examination. Collect organs and tissues for analysis, as well as ultrasound evaluation and cardiac transcriptome sequencing.

  Result: Slit2? The number of litters of Tg mice is significantly higher than that of C57BL/6J mice, and the high expression of Slit2 gene and protein can be detected. Slit 2? The spleen organ coefficient of Tg mice increased significantly, but no abnormal tissue structure was seen. Slit 2? Tg mice have significant changes in red blood cells, platelets, eosinophils, glucose, globulin, urea nitrogen, triglycerides, high-density lipoprotein, and atherosclerosis index. Except for the thickness of the anterior wall of the left ventricle at the end of diastole, the heart of Slit2? Tg mice did not differ significantly from other indicators. Of the 1,513 genes tested in heart tissue, 535 genes had a significant increase in expression. Or reduce, mainly involving 15 genes.

  Conclusion: Slit2? The biological parameters of Tg mice have been improved, making this model animal suitable for cardiovascular disease-related research.
