【Animal Modeling】-In situ hybridization of TRHmRNA at different developmental stages of Qinghai Lake naked carp embryo

  Purpose: This experiment aims to observe and analyze the expression of thyrotropin releasing hormone TRH gene in different stages of Qinghai Lake naked carp development.

  Method: Extract the total RNA of Qinghai Lake naked carp, prepare digoxin-labeled antisense TRH gene mRNA probe, and observe the expression of TRH gene by stereo microscope in situ hybridization.

  Results: Microscopic examination revealed that there was TRH gene positive hybridization signal in the diencephalon of Qinghai Lake naked carp embryos on the 6th to 8th day, and the embryonic diencephalon and midbrain on the 9th to 11th day. In the back of the brain, the medulla oblongata appears. Positive hybridization signals appeared for many days, and the expression level of diencephalon was higher than that of other brain tissues.

  Conclusion: In situ hybridization shows that THR gene is expressed in embryos of Qinghai Lake naked carp at 6-11 days, and TSH plays an important role in Qinghai Lake brain development and maturation.
