【Animal Modeling】-Distribution of Adenovirus Type 5 in C57BL/6J Mice After Different Routes of Infection

  Objective: To detect the distribution of adenovirus type 5 (Ad5) after C57BL6J infection by four methods.

  Method: 112C57BL/6J divide mice into 4 groups. Each mouse was injected with 100 μL of 1×109 pfu/mL Ad5 solution through nasogastric tube, gastric tube, tail vein, and abdominal cavity on the first 1, 2, 3, and 4 days after infection. , 5d, 6d, 7d each group killed 2 male and female mice, lymph nodes, testes, epididymis, uterus, ovary, spleen, kidney, liver, lung, heart, brain, blood and bone marrow tissues for fluorescence quantitative PCR.

  Result: Ad5 was detected in the lung and spinal cord tissue of the nose drip group, and Ad5 in the lung tissue was further reduced 4 days after infection. Ad5 was not detected in all tissues of the gastrointestinal feeding group. Detected in the liver and spleen. Ad5 was detected in the tail vein injection group, and the rate of decline was slower. Ad5 was detected in the spinal cord, spleen, epididymis and testis in the intraperitoneal injection group. The content of Ad5 in the spinal cord of the intraperitoneal injection group was about 10 times that of the nasal fluid group. The content of Ad5 in the spleen was similar to that in the tail vein injection group. Postnasal drip and testicular Ad5 only accounted for 1~about 10? of the content detected in other positive tissues.

  Conclusion: Ad5 lung infection experiment can be performed every 3 days, liver infection experiment can use tail vein injection, spleen infection experiment can use tail vein injection or intraperitoneal injection, spinal cord infection experiment can use intraperitoneal injection or internal injection method.
