【Animal Modeling】-The effect of Trichinella spiralis and its worm-derived protein on sepsis induced by cecal ligation and perforation in mice

OBJECTIVE: To observe the effect of caecal worm and its worm-derived protein on cecal ligation and perforation (CLP)-induced sepsis in mice.

Method: 80 male BALB/c mice were randomly divided into sham operation group, CLP group, Trichinella larvae (ML) pre-infection group, Trichinella soluble protein (SMP) treatment group, and excretion secretory protein (MES) group. . Treatment group. In the pre-ML infection group, 300 deltoid muscle larvae were orally infected 28 days before surgery, and the other group was intraperitoneally injected with PBS or SMP (25 μg/mouse) or MES (25 μg/mouse) 30 minutes after surgery. I injected it. The postoperative situation and 72-hour survival rate of the mice were observed. The serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), urea nitrogen (BUN) and creatinine (Cr) in the mouse serum were detected 12 hours after the operation. TNF-α, IL-6, IL-1β, IL-10 and TGF-β levels were observed in mouse liver and kidney tissues.

Result: Compared with the sham operation group, the 72-hour survival rate, ALT, AST, BUN, Cr serum levels and cytokines TNF-α, IL-6, IL-1β, and IL- of the CLP group were reduced. It is true. The levels of 10 and TGF-β were significantly increased (Pu003c0.05). The hepatic cords of the liver are arranged randomly, hepatocytes are edema, some of the renal vascular glomeruli contract, and renal tubular cells are edema. Compared with the CLP group, the serum ALT, AST, Cr, TNF-α and IL-1β levels in the pre-ML infection group decreased, and the IL-10 and TGF-β levels increased (Pu003c0.05). Serum ALTAST, Cr, TNF-α and IL-1β levels in the SMP treatment group decreased, and TGF-β levels increased (P\u003c0.05). The 72-hour survival rate of the MES treatment group increased significantly. , And serum ALT, AST, BUN, Cr, TNF-α, IL-6 and IL-1β levels were significantly reduced, and IL-10 and TGF-β levels were significantly increased (Pu003c0.05). , The structural damage to liver and kidney tissues is significantly reduced.

Conclusion: Trichinella spiralis and its worm-derived proteins can reduce the release of pro-inflammatory factors in the serum of CLP-induced sepsis mice, and promote the release of immune regulatory factors. Among them, the role of MES is more important, which can reduce the damage to the structure and function of liver and kidney.
