【Animal Modeling】-Experimental Study on the Differentiation of Bone Marrow Mesenchymal Stem Cells into Cardiomyocyte-like Cells Induced by Jiawei Danshen Decoction in Vitro

  Objective: To investigate the effect of Jiawei Danshen Decoction on the differentiation of bone Messechymal stem cells (BMSC) into cardiomyocyte-like cells in vitro.

  Method: Obtain rat BMSCs by adherence method, and detect cell surface antigens by flow cytometry. MTT method was used to detect the maximum non-toxic concentration of Jiawei Danshen Decoction to BMSC. It was induced by the third generation cells (normal control group, Jiawei Danshen Decoction group, 5-azacitidine group, Jiawei Danshen Decoction + 5-azacitidine group). Western blot detection of cell binding protein (desmin), α-myosin heavy chain (α-MHC), cardiac troponin T (cTnT) protein expression in each group, RT-PCR detection of GATA-4, gap expression in each group Conexin 43 (CX43) and kx2-5 mRNA.

  Result: The positive rate of CD90 on the surface of rat bone marrow mesenchymal stem cells is higher than 95? The positive rate of CD34 and CD45 is lower than 5? The maximum non-toxic concentration of flavored salvia beverage in BMSC is 0.625g/L. The expression of desmin, α-MHC and cTnT protein in the Jiawei Danshen Decoction group, 5-azacitidine group, and the Jiawei Danshen Decoction + 5-azacitidine group were up-regulated compared with the normal control group, and the expression of GATA-4, CX43, Nkx2 -5 mRNA The level was up-regulated, and the difference was statistically significant (Pu003c0.01). Compared with the Jiawei Danshen Decoction group or 5-azacytidine group, GATA-4, CX43 and kx2-5, the expression of Desmin, α-MHC and cTnT protein in the Jiawei Danshen Decoction + 5-azacytidine group was up-regulated. mRNA was up-regulated and the difference was statistically significant (Pu003c0.01).

  Conclusion: Jiawei Danshen Decoction can induce BMSC to transform into cardiomyocyte-like cells, and it has a synergistic effect when combined with 5-azacitidine.
