【Animal Modeling】-The effect of androgen deficiency on visceral fat accumulation and inflammatory gene expression in miniature pigs under high-fat environment

Objective: To study the effects of androgen deficiency on visceral fat deposition, serum hormones and inflammation-related gene expression in miniature pigs in a high-fat environment.

Method: Sexually mature male Wuzhishan mini pigs were randomly divided into 3 groups: uncastrated (SHAM), castrated (CAS), castrated plus testosterone (CAS+T), 6 in each group. I have a pig. Animals are fed a high-fat diet. At the end of the 12-week experiment, blood was drawn to detect changes in serum hormone levels, visceral fat was separated and weighed, and fluorescent quantitative PCR technology was used to generate lipolysis and lipolysis and inflammation-related genes to detect the expression. ..

Results: (1) Castration significantly reduced the serum testosterone content of high-fat edible miniature pigs, but increased the serum leptin content. Exogenous testosterone treatment can restore serum testosterone and leptin levels in castrated miniature pigs. The visceral fat of high-fat diet miniature pigs is deposited in large amounts in the high-fat diet. After testosterone treatment, the visceral fat content of castrated miniature pigs was significantly reduced. (3) Castration and testosterone treatment are affected. Fat metabolism genes such as FAS, ACC, HSL and visceral adipose tissue ATGL had no significant effect on the expression of high-fat diet miniature pigs; CCL23 and SAA in the visceral adipose tissue of high-fat diet miniature pigs, as well as exogenous testosterone For the treatment of castrated miniature pigs. Pigs can reduce the expression of these genes.

Conclusion: Castration-induced androgen deficiency promotes visceral fat deposition in high-fat diet miniature pigs and up-regulates the expression of inflammatory genes in visceral adipose tissue. Treatment with testosterone can improve visceral fat accumulation and inflammation in castrated miniature pigs.
