【Animal Modeling】-The effect of self-made Gehua Jiejiu Dizhi Decoction on liver fat deposition and PXR expression in alcoholic fatty liver mice

  Objective: To understand the effect of homemade Gehua Jiejiu Dizhi Decoction on liver fat deposition in alcoholic fatty liver (AFLD) mice, and observe its effect on liver tissue pregnane receptor (PXR) and its regulatory target genes CYP3A11 and CYP3A25 protein mRNA expression.

  Methods: 29 male C57BL/6J mice were randomly divided into normal group (5), model group (8), high-dose intervention group (8), and low-dose intervention group (8). The AFLD mouse model was adopted The National Institutes of Health's Alcohol Abuse and Alcoholism (NIAAA) method was created to take high and low doses of Pueraria lobata Relief Decoction for 9 consecutive days. Enzyme immunoassay was used to detect serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels, and histopathological oil red O staining was used to detect liver and gallbladder deposits. Use real-time fluorescent quantitative PCR and immunohistochemical methods to detect PXR. , CYP3A11, CYP3A25 mRNA and PXR protein expression.

  Result: Compared with the model group, the liver fat deposition of the intervention group was significantly reduced in a dose-dependent manner, and the expression levels of PXR and CYP3A25 were significantly increased in the model group and the normal group (P). u003c0.01); The serum ALT level of mice in the model group was significantly increased (Pu003c0.01), and the other groups were also the same; there was no significant difference in CYP3A11 mRNA transcription levels in each group (P≥0.05)).

  Conclusion: Homemade Gehua Jiejiu Dizhi Decoction has a significant therapeutic effect on AFLD in experimental mice. This may be related to the up-regulation of PXR and its target gene CYP3A25.
