【Animal Modeling】-High-throughput sequencing method to study the effect of different pH drinking water on the gut microbes of SPF mice

  Objective: To study the effect of drinking water with different pH values on the gut microbial diversity of SPF mice.

  Method: Ninety 21-day-old SPF mice were randomly divided into 3 groups, 30 in each group, acidic water (pH3.0), neutral water (pH7.0), and alkaline water. Running with water (pH 9.0), the breeding period is 3 months. On the 4th, 8th and 12th weekends, 10 animals per group died. The serum was collected to detect bacteria, viruses and parasites according to the SPF national standard, and the blind content was obtained. I designed the primer. According to the maintainability of bacterial 16SRNA, build a library, perform high-throughput (Illumina) sequence, obtain 10G data, and perform data statistics and cluster analysis.

  Result: The amount of intestinal flora of mice treated with pH 3.0 is moderate and the stability is high.

  Conclusion: Under the conditions of large-scale reproduction, drinking water mice with pH 3.0 can maintain the diversity of intestinal microbes for a long time, and the stability tends to be stable. This research provides theoretical and technical support for production practice to maintain stable animal quality for a long time, and is also a specific application of gastrointestinal metagenomics technology in the field of animal quality control.
