[Animal modeling]-Flow cytometry analysis of the phenotypic characteristics of PMA-induced THP-1 differentiation into macrophages

  Purpose: To use flow cytometry to reveal that THP-1 cells only differentiate into macrophages induced by PMA.

  Method: First use PMA to stimulate THP-1 cells to differentiate into macrophages, and then use LPS, IFN-γ, IL-6 to induce cells to differentiate into M1 macrophages, I will. Induces cells to produce IL-4, IL-13 and IL-6 M2 type macrophages. The morphological changes of the three types of cells were observed under a microscope, and the differences in the expression of the main CD molecules of each cell were detected by flow cytometry.

  Result: After THP-1 differentiates into macrophages, adherent growth occurs, and the cell morphology is relatively regular round or oval. M1 and M2 cells are irregular, with obvious cell protrusions and large cell size. The expression of CD molecules on the surface of macrophages related to the antigen presentation function is low, and most of them are negative, but the expression of these CD molecules on the surface of M1 and M2 cells is relatively high.

  Conclusion: Macrophages differentiated from THP-1 cells are only stimulated by PMA, their antigen presentation ability is weak, and they are basically in a dormant state.
