  OBJECTIVE: To compare mouse models of delayed constipation induced by loperamide hydrochloride, compare the differences in the models induced by different doses and different administration times, and establish a more stable mouse model of delayed constipation induced by loperamide hydrochloride .

  "Method: Use healthy male C57BL/6 mice to prepare 0.025, 0.25, and 2.5 mg/mL loperamide hydrochloride suspensions by gastric administration. The administration time is twice a day at 9 am and 3 pm. The effect of the model was evaluated from the detection of intestinal propulsion rate and stool volume, and the effects of different doses and different administration times on the slow transit constipation mouse model were observed.

  Results: 1) Compare the effects of different doses of loperamide hydrochloride in the concentration range of 0.5mg/kg~50mg/kg on the model. By comparing the intestinal propulsion rate between the groups, the result is 50mg, which shows that the /kg group comparison, The intestinal advancement rate and defecation volume of the mice in the blank group were significantly reduced, and the difference was statistically significant (P\u003c0.05).

  Conclusion: The study found that continuous gavage at a dose of 50 mg/kg for 7 days can significantly reduce the rate of intestinal progression in mice and significantly reduce the amount of defecation. This allows you to create a relatively stable mouse model of impaired colonic transport.
