【Animal Modeling】-Establishment and evaluation of murine sepsis model

  Objective: To establish and evaluate a mouse sepsis model, to study the pathogenesis of sepsis, and to provide model animals for the development of anti-inflammatory drugs.

  Method: Peel and puncture (CLP) induce sepsis in mice through animal survival rate, postoperative bacterial load, blood routine and blood biochemical indicators, cytokine levels, histopathological changes, etc. The model is evaluated.

  Result: The mortality of mice is closely related to the site of cecal ligation. The survival rate of 50? cecal ligation mice at 12 days is about 40? 4 days after ligation, 75? mice died (Pu003c0.01). Compared with the sham operation group, the blood and intraperitoneal bacterial levels of 50-strand CLP mice increased, and the white blood cells decreased. The difference is significant (Pu003c0.001). The levels of liver transaminase ALT, AST and serum urea nitrogen BUN in CLP mice increased with significant difference (Pu003c0.01), and the levels of inflammatory factors IL1α, IL6, IL10, MIP1α, MIP1β, and TNFα increased. 48 hours after the operation, the liver and lungs of the mice showed obvious histopathological damage.

  Conclusion: The mouse CLP model has typical pathological features of sepsis, which provides a better animal model for subsequent anti-inflammatory drug research.
