【Animal Modeling】-Why build a ZDF rat model of diabetes and coronary heart disease?

  Objective: To construct a rat model of type 2 diabetes with coronary heart disease.

  Method: The Zucker diabetic fat rat (Zuckerdiabeticfatty, ZDF) model of type 2 diabetes and coronary heart disease was prepared by feeding a high-fat diet combined with isoproterenol hydrochloride (ISO) continuous injection. The model was evaluated by serum creatine kinase (CK), serum creatine kinase isoenzyme (CK-MB), ECG ST, and cardiac tissue morphology.

  Result: The expression of CK in the non-DM model group and the DM-CHD group gradually increased with the increase in the number of administrations. The expression of CK-MB first increased and then decreased. The rats developed myocardial injury. I did it. The deviation of the ST segment of the Read II ECG of model rats indicates the formation of myocardial injury. The results of myocardial pathological section of rats in the injection model group showed that the myocardial cells of the blank control group were normal; the area of myocardial necrosis in ZDF rats was 3 times that of myocardial sections after continuous ISO injection, and 1/2 of myocardial sections after 5 days. ,Four.

  Conclusion: Changes in CK and CK-MB content, changes in ECG ST waves, and myocardial pathological slices all indicate the success of the model. ISO can cause a certain amount of myocardial damage, and continuous infusion of ISO can be used to establish a coronary heart disease model. In this article, we have established a simple method for constructing a rat model of diabetes and coronary heart disease with a high-fat diet combined with continuous ISO injection.
