【Animal Modeling】-Isoproterenol-induced changes in the pathological tissue of myocardial cells caused by specific myocardial ischemia in rats

Objective: To observe the expression of HE staining in myocardial tissue slices caused by subcutaneous injection of isoproterenol for different days (1 mg/kg body weight). VEGF (vascular endothelial growth factor) and FCF (fibroblast growth factor) cause a large The electrocardiogram wave of the changes in mouse-specific cardiomyopathy.

Method: Except for the blank group, rats in each group were injected subcutaneously with isoproterenol (1mg/kg) for continuous subcutaneous injection for 2 days and for the continuous group with isoproterenol (1mg/kg) multipoint subcutaneous injection. .Multiple injections under the skin. Continuous subcutaneous injection of furan propionic acid (1mg/kg) for 4d group and isoproterenol (1mg/kg) for 6d group. One week after the administration, the rats were anesthetized, the electrocardiogram was taken, and the heart was sampled. HE staining was used to observe the changes of rat myocardial cell injury, and immunohistochemical staining to observe VEGF (vascular). Changes in the expression of endothelial growth factor) and FGF (saturated fibroblast growth factor).

Results: Continuous subcutaneous injection of isoproterenol (1mg/kg) for 6 days showed significant differences in HE stained tissue sections under the microscope compared with the blank control group. Myocardial tissue sections in the model group were VEGF (vascular endothelial growth factor), FCF ( Fibroblast Growth Factor) expression levels are also significantly different from the blank control group.

CONCLUSION: The results of this study indicate that HE staining of myocardial tissue slices of specific cardiomyopathy in rats in the isoprenaline (1mg/kg) 6d group, continuous subcutaneous VECF (Vascular Endothelial Growth Factor) factor compared with the blank control group, indicates that injection . Due to the significant difference between FGF (fibroblast growth factor) expression and ECG T wave changes, the efficacy of specific cardiomyopathy should be evaluated by subcutaneous injection of isoproterenol (1 mg/kg) for 6 consecutive days.
