[Animal modeling]-Intervention of electroacupuncture on activation of dorsal root ganglion satellite glial cells and P2X7 receptor expression in rats with inflammatory pain

  OBJECTIVE: To observe the effects of electroacupuncture on the mechanical pain threshold (foot withdrawal threshold, PWT) of the dorsal root ganglion (DRG) of rats with chronic inflammatory pain and activation of satellite glial cells and P2X7 receptors. To explore the influence of electroacupuncture on the peripheral mechanism of inflammatory pain in rats.

  Method: Part 1: Rats are completely random blank group (Congroup, n=12), inflammatory pain group (CFAgroup, n=12), inflammatory pain + electroacupuncture group (CFA+EAgroup, n)=12, )), Yantong+sham electroacupuncture group (CFA+sEA group, n=12). Rats with inflammatory pain were injected with 0.1 mL of complete Freund’s adjuvant (CFA) subcutaneously in the center of the palm of the right hind paw to establish a model. Acupuncture points "Zusanli" and "Kunrun" electroacupuncture and electroacupuncture parameters. Density wave, frequency 2/100Hz, intensity 0.5-1-1.5mA (each intensity treatment is 10 minutes), once a day for 7 days. Before and after modeling and 1, 3, 7, 8, 10, 12, and 14 days after modeling, the mechanical pain threshold of rats in each group was tested, and the glial acidity of the affected DRG was tested. I did it. The rats in each group were detected by Western blotting 14 days after modeling. Activate protein (glial fibrillary acidic protein, GFAP) and P2X7 receptors. Part 2: Rat inflammation pain + DMSO group (CFA+DMSO group, n=8), inflammation pain + P2X7R inhibitor A740003 group (CFA+A740003 group, n=10), inflammation pain + electroacupuncture + salt (CFA)+electroacupuncture+NS group, n=8), inflammatory pain+electroacupuncture+P2X7R agonist BzATP group (CFA+electroacupuncture+BzATP group, n=12), respectively used as anti-inflammatory drugs. The changes in rat PWT were detected before surgery, before modeling, after modeling, and after administration.

  Results: (1) The pain threshold of CFA-induced inflammation model rats was significantly reduced (P0.05). (2) Compared with the CFA+DMSO group, intrathecal injection of the P2X7 inhibitor A740003 significantly increased the mechanical pain threshold of CFA rats (Pu003c0.01); the PWT of the CFA+EA+BzATP group was significantly lower than that of the control group CFA+ PWT in. EA+NS group (Pu003c0.01).

  Conclusion: Activation of rat dorsal root ganglion satellite glial cells and inhibition of P2X7 receptor expression may be one of the peripheral mechanisms of electroacupuncture analgesia involved in the treatment of chronic inflammatory pain by electroacupuncture.
